Description

Introduction: Beta-catenin (or beta-catenin) is a protein that in humans is encoded by the CTNNB1 gene. In Drosophila, the homologous protein is called armadillo. beta-catenin is a subunit of the cadherin protein complex and has been implicated as an integral component in the Wnt signaling pathway. When beta-catenin was sequenced it was found to be a member of the armadillo family of proteins. These proteins have multiple copies of the so-called armadillo repeat domain which is specialized for protein-protein binding. When beta-catenin is not associated with cadherins and alpha-catenin, it can interact with other proteins such as ICAT and APC.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to beta-Cat. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for beta-Cat and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then substrate solution is added to each well. Only those wells that contain beta-Cat, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of beta-Cat in the samples is then determined by comparing the O.D. of the samples to the standard curve
